Rapid and Flexible cfDNA Extraction Kit

This product enables rapid and accurate extraction of cfDNA from plasma. It utilizes an easy-to-use spin column, eliminating the need for vacuum filtration. After spinning, DNA is efficiently recovered with a minimal elution volume of just 35 μL, allowing more cfDNA to be used in subsequent analysis steps, such as ddPCR and NGS, thereby enhancing the sensitivity of variant detection.

Optimized Efficient Workflow

The kit offers a flexible workflow that allows for easy processing of up to 10 mL of biological samples in under 90 minutes. It enables efficient extraction of cfDNA without the use of carrier RNA, ensuring accurate quantification and library preparation of cfDNA. No special equipment, such as magnets or vacuum devices, is required.

Validated Kit

To evaluate the performance of Cell3™ Xtract, a comparison was conducted with four existing manual cfDNA extraction kits (from companies Q, A, O, and M). These kits included both spin column and bead-based methods. The performance evaluation was carried out using plasma samples from a pregnant woman carrying a male fetus.

Comparison to Spin Column Based Kits

To compare Cell3™ Xtract with other kits, cfDNA was extracted from 1 mL of plasma from the same patient following each company’s protocol. According to quantitative PCR data, the Cell3™ Xtract kit demonstrated performance equivalent to Company Q’s kit and outperformed Company A’s kit. In Company Q’s kit, the use of carrier RNA resulted in a DNA concentration measured approximately 50 times higher, and fragment analysis revealed a high molecular weight peak (Figure 1).

Figure 1 CfDNA was extracted from 1 ml of plasma for each kit: the same sample from a male pregnancy at 14 weeks gestation was used. Quantitative PCR evaluation was conducted to detect and compare the extracted fetal cfDNA portion using the Cell3™ Direct: Fetal Sex Determination kit (Nonacus), which includes chromosome Y specific targets (TSPY, SRY, DAZ) and a control target (CCR5). Fragment size distribution was measured using high sensitivity reagents on the Tapestation 2200 (Agilent). DNA concentration was determined using high sensitivity reagents on the Qubit 3.0 (Invitrogen).

Comparison to Bead Based Kits

Based on quantitative PCR data and DNA concentration measurements, Cell3™ Xtract outperformed Company M’s kit and delivered comparable results to Company O’s kit (Figure 2).
